چکیده

As β-glucosidases represent the major bottleneck for industrial degradation of plant biomass, great efforts are being devoted to discover both novel and robust versions these enzymes, as well develop efficient inexpensive ways produce them. In this work, raw glycerol from chemical production biodiesel was tested carbon source fungus Talaromyces amestolkiae with aim producing enzyme β-glucosidase-enriched cocktails. Approximately 11 U/mL β-glucosidase detected in cultures, constituting cellulolytic activity. Proteomic analysis showed BGL-3 most abundant protein main β-glucosidase. This crude successfully used supplement a basal commercial cocktail (Celluclast 1.5 L) saccharification pretreated wheat straw, corroborating that even hardly exploitable wastes, such glycerol, can be secondary materials valuable enzymatic preparations framework circular economy.

Direct glucose production from lignocellulose using Clostridium thermocellum cultures supplemented with a thermostable β-glucosidase

BACKGROUND Cellulases continue to be one of the major costs associated with the lignocellulose hydrolysis process. Clostridium thermocellum is an anaerobic, thermophilic, cellulolytic bacterium that produces cellulosomes capable of efficiently degrading plant cell walls. The end-product cellobiose, however, inhibits degradation. To maximize the cellulolytic ability of C. thermocellum, it is imp...

Differential β-glucosidase expression as a function of carbon source availability in Talaromyces amestolkiae: a genomic and proteomic approach

BACKGROUND Genomic and proteomic analysis are potent tools for metabolic characterization of microorganisms. Although cellulose usually triggers cellulase production in cellulolytic fungi, the secretion of the different enzymes involved in polymer conversion is subjected to different factors, depending on growth conditions. These enzymes are key factors in biomass exploitation for second genera...

Screening, production, optimization and characterization of β-glucosidase using microbes from shellfish waste

An extracellular β-glucosidase was isolated from Proteus mirabilis VIT117 found to be growing on prawn shells. The enzyme production was found to be enhanced (14.58 U/ml) when the culture was maintained at pH 9 and provided with sorbitol as carbon source, yeast extract as nitrogen source and incubated at 37 °C for approximately 72 h. Statistical methods like Plackett-Burman and RSM were also ap...

A novel, highly efficient β-glucosidase with a cellulose-binding domain: characterization and properties of native and recombinant proteins

Background Cellulose, the most abundant biopolymer on earth, is an alternative for fossil fuels as a renewable feedstock for the production of second-generation biofuels and other chemicals. The discovery of novel, highly efficient β-glucosidases remains as one of the major bottlenecks for cellulose degradation. In this context, the ascomycete Talaromyces amestolkiae, isolated from cereal sampl...

Evolution of D-lactate dehydrogenase activity from glycerol dehydrogenase and its utility for D-lactate production from lignocellulose.

Lactic acid, an attractive, renewable chemical for production of biobased plastics (polylactic acid, PLA), is currently commercially produced from food-based sources of sugar. Pure optical isomers of lactate needed for PLA are typically produced by microbial fermentation of sugars at temperatures below 40 °C. Bacillus coagulans produces L(+)-lactate as a primary fermentation product and grows o...
